Verdict: Giving this to Spidey. Tom Holland’s Spider-Man in his youth gave each of them a good fight in of himself, even beating Bucky. Neither one of them is gonna be happy tangling with not only a far more experienced and far stronger Spider-Man, but one who’s also a lot stronger than normal—and borderline bloodlusted at that.

Hitting the Shield will send the black suit reeling leaving him open for Bucky to get hits in.

^ They wouldn’t know to do that, tho. That requires them to understand the Symbiote’s weakness, when I assume both parties are going into this blind.

Which is why I’m leaning towards Spider-Man, too fast and strong for them to overcome. Cap’s a beast, but stopping a helicopter is nothing compared to stopping a runaway train, and neither Cap or Bucky have the feats to suggest they could continue fighting after getting sent through multiple walls by a point-blank bomb going off (generally Super Soldier tier characters are in rough shape after being near explosions that don’t crash them through brick walls).
